We measured several characteristics of water molecules and imidazoles in the MD simulations (table S4). The number of water molecules within the membrane region is similar for the R-HC8 and S-HC8 constructs, 68 and 65, respectively, and is significantly higher than for the HC8 system featuring 39 water molecules. This results in the formation of water wires of roughly 3.5 nm in length, much longer than the length of ~1.0 nm of the water wires observed in the selectivity filter of AQP (22). We previously characterized the structure of I-quartet channels in the membrane region and concluded that the most representative channels in our simulation setup are the two central ones, which are less perturbed by boundary effects than the more exposed lateral ones (14). Hence, most of the simulation analysis focuses on these two central channels depicted in Fig. 4A. The visually apparent water ordering and orientation is in part due to anchoring sites provided by the imidazole structure across the membrane. Another contribution to water structuring originates in the electrostatic field generated by the ensemble of charges in the I-quartet system as depicted in Fig. 4B. A positive to negative potential field gradient builds up from bottom to top, driven by an additive effect of the orientation and ordering of imidazole dipoles, which seems to be highly correlated with the membrane contractions observed from QCM-D experiments. Qualitatively, this effect seems proportional to the preserved ordering of the imidazoles and decreases from R-HC8 to S-HC8 and HC8.

Genetic analyses dependent on data from modern populations do not bear directly on the geographic locations of the divergence events (13). For instance, the data used to generate the Beringian Incubation Model only require that Native American ancestors were geographically isolated from wherever the East Asian/Siberian ancestors inhabited during the time of isolation (15). The genetic data do not require that this isolation occurred in central or eastern Beringia—it could range across a vast area, from Cis-Baikal to Hokkaido. So far, complete nuclear genomes from ancient samples produced in recent years (16–18) are not able either to substantially narrow the divergence estimate or to clarify spatial routes of the initial peopling.

Moreover, the SERS effect of M, T, MT, MG, MTG, cMGT, and MGT substrates to CuPc molecules was compared to further confirm the contribution of the enrichment of CuPc on MGT by porous TiO2 to the SERS effect (Fig. 4K). We observed no significant enhancement on M, T, and MT, which indicated that GO mainly contributed to the SERS effect. On the other hand, compared with MGT, MG and MTG also have multilayered GO nanosheets. However, the EF values of cMGT (4.67 × 105), MTG (1.06 × 106), and MG (3.05 × 105) were lower than that of MGT for CuPc (8.08 × 106; Table 1). Significantly, the enhancement effect of cMGT is much lower than that of MGT (about 51% Raman intensity for CuPc on MGT), which was attributed to cMGT preventing the interaction between GO and CuPc. Consequently, compared with MTG, cMGT, and MG, the significant SERS effect of MGT should be ascribed to the enrichment of CuPc on the porous TiO2 shell for MGT. Consequently, the remarkable enhanced EF toward CuPc molecules obtained at MGT substrates should be attributed to our designed ternary heterostructure: (i) the selective SERS effect of GO toward CuPc, (ii) the resonance effect of CuPc under 633-nm laser excitation, and (iii) CT between GO and TiO2 that remarkably enhances the Raman signal of CuPc and the enrichment of CuPc molecules by the porous TiO2 shell of MGT.
